Arachidonate 15-lipoxygenase-1 (15-LOX-1, encoded by the ALOX15 gene) is a non-heme iron-containing dioxygenase enzyme that catalyzes the stereo-specific peroxidation of polyunsaturated fatty acids such as linoleic and arachidonic acids to generate lipid mediators including 15-hydroperoxyeicosatetraenoic acid (15-HETE) and 13-hydroxyoctadecadienoic acid (13-HODE). It plays a prominent role in cell differentiation, membrane remodeling, and inflammation resolution, notably contributing to the non-immunogenic removal of apoptotic cells and the generation of specialized pro-resolving mediators in macrophages. Dysregulated ALOX15/15-LOX-1 activity is implicated in various pathologies, including cancer (particularly in tumor genesis and progression), inflammation, cardiovascular, and neurodegenerative disorders. Inhibitors of 15-LOX-1 are being explored as potential therapeutics, especially to interfere with regulated cell death and pathological inflammation.
Inhibitors of 15-LOX-1 block the formation of lipid peroxides, reducing oxidative stress and interfering with programmed cell death pathways involved in inflammation and disease.
