Overview

Arachidonate 15-lipoxygenase type B (ALOX15B) is an iron-dependent enzyme of the lipoxygenase family that catalyzes the stereo- and regio-specific peroxidation of polyunsaturated fatty acids such as arachidonic acid, eicosapentaenoic acid, docosahexaenoic acid, and linoleic acid. Its primary activity is the oxygenation of arachidonic acid at carbon 15, leading to the generation of 15-hydroperoxy derivatives (such as 15-HpETE) and downstream lipid mediators with both pro- and anti-inflammatory effects. ALOX15B is constitutively expressed in human macrophages and is involved in cholesterol homeostasis, cellular oxidative processes, and the formation of specialized pro-resolving mediators. Its altered expression is implicated in a variety of diseases, including cancer, atherosclerosis, and inflammatory conditions. The precise role of ALOX15B in human health and disease is complex and context-dependent, with both potential tumor-suppressive and tumor-promoting effects reported depending on tissue type and pathological state.
