Overview

Arachidonic acid pathway inhibitors are a class of molecules that target key enzymes or receptors involved in the metabolism of arachidonic acid (AA). The AA pathway is central to the production of bioactive lipid mediators, including prostaglandins, leukotrienes, and thromboxanes—collectively known as eicosanoids—which play critical roles in inflammation, immunity, cardiovascular function, and cancer progression. The main targets for inhibition include Phospholipase A2 (PLA2), Cyclooxygenases (COX-1 and COX-2), and Lipoxygenases (LOXs). These inhibitors work by blocking one or more steps in the enzymatic conversion of membrane phospholipids to pro-inflammatory eicosanoids, resulting in reduced production of inflammatory mediators responsible for pain, fever, swelling, bronchoconstriction, platelet aggregation, tumor growth promotion, and other pathophysiological processes. They are used or investigated for management of pain and inflammation, treatment/prevention of cardiovascular diseases, asthma management, cancer therapy adjuncts, and autoimmune disorders. Recent research focuses on developing selective enzyme isoform inhibitors, exploring natural product-derived compounds, and investigating combination regimens with existing therapies.
