The arginine biosynthesis pathway is a multi-step metabolic route that synthesizes arginine, an amino acid, from precursor molecules including citrulline, via key enzymes such as argininosuccinate synthase and argininosuccinate lyase. This pathway is essential for cellular nitrogen metabolism, urea cycle function, and polyamine synthesis. In mammals, the pathway is most active in the kidneys and intestines. Arginine biosynthesis is essential for cell growth and proliferation, and deficiency can result in metabolic disorders and increased susceptibility to some cancers. Therapeutically, several enzymes within the pathway are targeted by drugs, especially in cancers where the pathway is dysregulated or silenced, rendering tumors dependent on external arginine. In pathogens such as fungi, the pathway contributes to virulence and is a promising target for antifungal drug development. The pathway is regulated at genetic, biochemical, and epigenetic levels, and includes complex feedback and feedforward interactions with nutrient and stress responses.
