The arginine vasopressin V2 receptor (AVP-V2R) is a G protein-coupled receptor primarily located in the kidney collecting ducts. It mediates the antidiuretic effects of arginine vasopressin (AVP), regulating water reabsorption via aquaporin 2 trafficking. Activation increases cAMP, leading to PKA activation and AQP2 translocation to the apical membrane. Mutations in the AVPR2 gene cause nephrogenic diabetes insipidus. The receptor is also implicated in vascular endothelial function and is a therapeutic target for hyponatremia and coagulation disorders.
Agonism or antagonism of the AVP-V2R, leading to changes in cAMP/PKA signaling and downstream effects on water reabsorption and coagulation factor release.
