ARHGEF35 antisense RNA 1 (ARHGEF35-AS1, also called DDGC) is a long non-coding antisense RNA implicated in the regulation of DNA repair and differentiation within ovarian granulosa cells. It exerts its effect by stabilizing RAD51 messenger RNA—critical for homologous recombination DNA repair—through competition for miR-589-5p binding, functioning as a competitive endogenous RNA (ceRNA). Additionally, DDGC modulates granulosa cell differentiation by facilitating interaction between the chaperone protein HSP90 and the transcription factor WT1, thereby protecting WT1 from ubiquitin-mediated degradation and allowing proper cell differentiation and hormone production. Aberrant expression or silencing of ARHGEF35-AS1 leads to decreased DNA repair capacity, abnormal cell differentiation, and has been linked to premature ovarian insufficiency. To date, there are no drugs or diagnostic agents targeting ARHGEF35-AS1.
