Armadillo-like helical domain-containing protein 3 (ARMH3) is a highly conserved, ubiquitously expressed cytoplasmic protein localized to the Golgi complex. It binds the small GTPase ARL5, acting as its effector, and is essential for recruiting and activating phosphatidylinositol 4-kinase IIIβ (PI4KB), the main generator of PI4P lipids at the trans-Golgi network. ARMH3 thereby contributes to Golgi function—specifically, the regulation of glycan modifications and maintenance of Golgi structure—by supporting recruitment of PI4P-binding proteins such as the oncoprotein GOLPH3 and glycan-modifying enzymes. It is predicted to be predominantly α-helical with a coiled-coil and potential protein-protein interaction motifs, but lacks known enzymatic or receptor activity. Despite some structural and functional characterization, ARMH3 is not currently a therapeutic target, and there are no approved drugs, known biomarkers, or major safety concerns associated with its modulation.
