Overview

Armadillo repeat-containing protein 10 (ARMC10) is a mitochondrial membrane-associated protein characterized by armadillo repeats—protein motifs involved in mediating protein-protein interactions[1][5]. ARMC10 functions as an effector of AMP-activated protein kinase (AMPK) and is directly phosphorylated at serine 45 by AMPK, a modification that regulates mitochondrial fission; ARMC10 overexpression promotes fission, while loss prevents AMPK-driven changes[1]. ARMC10 is also involved in mitochondrial trafficking by interacting with the KIF5/Miro1-2/Trak2 complex, particularly in neurons, and helps prevent amyloid-beta-induced mitochondrial fragmentation and associated neurotoxicity[3][4][5]. Experimental studies show that ARMC10 can regulate neural precursor cell proliferation and may act as a negative regulator of the Wnt/β-catenin signaling pathway, possibly contributing to both nervous system development and suppression of tumorigenesis[4]. Although variants have been linked to hepatocarcinogenesis, ARMC10 itself is not widely validated as a therapeutic target, and there are no known approved drugs or biomarkers directly targeting or using ARMC10[1][3][5].
