Armadillo repeat-containing protein 12 (ARMC12) is a multifunctional protein belonging to the armadillo (ARM) repeat protein family, characterized by tandem repeats facilitating diverse protein-protein interactions[4]. ARMC12 plays critical roles in both cancer biology and reproduction. In neuroblastoma, ARMC12 acts as an oncogene by binding to the protein RBBP4, thereby promoting formation and enzymatic activity of polycomb repressive complex 2, which suppresses transcription of tumor suppressor genes and drives cell proliferation and aggressiveness[1][4]. Elevated ARMC12 expression is associated with poor prognosis in neuroblastoma and possibly other cancers. In the male reproductive system, ARMC12 is required for proper assembly of the mitochondrial sheath in the sperm midpiece, an essential process for sperm motility and fertility; loss of ARMC12 causes disrupted mitochondrial organization, impaired sperm motility (asthenozoospermia), and infertility[2][3][5][6]. ARMC12 functions as a mitochondrial peripheral membrane protein, interacting with components such as VDAC2, VDAC3, MIC60, TBC1D21, and others to mediate mitochondrial dynamics and membrane adherence[2][5][6]. Consequently, ARMC12 is a candidate therapeutic target in cancer and a biomarker in certain reproductive and developmental contexts, but tissue selectivity and fertility preservation are important safety considerations[4].
In cancer: ARMC12 facilitates formation and activity of polycomb repressive complex 2 (PRC2) through interaction with retinoblastoma binding protein 4 (RBBP4), contributing to transcriptional repression of tumor suppressor genes In reproduction: Acts as a scaffold/adherence factor for mitochondrial organization via interactions with mitochondrial proteins (e.g., VDAC2, VDAC3, MIC60, TBC1D21) during spermiogenesis
