Overview

ARMC2 encodes a structural protein (~97 kDa in humans) characterized by armadillo (ARM) repeats that form superhelical platforms for protein-protein interaction[3]. The protein is highly conserved among species with motile cilia and flagella, and is essential for the proper assembly and stability of the axonemal central pair complex (CPC), a key microtubule structure in sperm flagellum[1][2][3]. Bi-allelic mutations in ARMC2 are causative of severe sperm flagellum malformations, resulting in male infertility both in humans and animal models[1][2]. In organisms like Chlamydomonas, ARMC2 acts as a cargo adapter for intraflagellar transport (IFT), essential for bringing radial spoke proteins into developing flagella[2]. ARMC2 is not currently a drug target and does not have established biomarkers or associated safety concerns, but it may be implicated in ciliopathies and infertility diagnostics[1][2][4].
