Overview

Arrestin domain-containing protein 5 (ARRDC5) is an alpha-arrestin family adapter protein involved in the regulation of protein transport and endocytosis, as well as spermatogenesis in mammals[1][3][4][5]. It is expressed in testicular germ cells, particularly during the round and elongated spermatid phases, and its loss leads to male sterility without affecting females, establishing it as an essential regulator of sperm maturation[4][5]. ARRDC5 is also implicated in osteoclast differentiation by regulating V-type ATPase localization and bone resorption activity[2]. The protein has been identified as a potential target for non-hormonal male contraception and as a possible prognostic marker in certain cancers, such as lung adenocarcinoma, although its molecular mechanism of action, especially in human disease, is not fully defined[1][3][4].

Mechanism of action
