Art v 1-specific Immunoglobulin E (IgE) is a specialized antibody produced by the immune system in response to exposure to Art v 1, the major allergen of Artemisia vulgaris (mugwort) pollen (Source: WHO/IUIS Allergen Nomenclature Sub-Committee). These antibodies play a central role in the pathogenesis of mugwort-induced allergic diseases, such as seasonal allergic rhinitis and asthma (Source: PMID: 11339815). Upon exposure to mugwort pollen, Art v 1-specific IgE molecules bound to high-affinity FcεRI receptors on the surface of mast cells and basophils are cross-linked by the Art v 1 allergen (Source: UniProt P80176). This cross-linking triggers the degranulation of these effector cells, leading to the release of inflammatory mediators like histamine, which cause the clinical symptoms of allergy (Source: StatPearls, Type I Hypersensitivity). In therapeutic contexts, IgE is a target for monoclonal antibodies like Omalizumab, which sequester free IgE and prevent its binding to receptors (Source: FDA Label, Xolair). Additionally, allergen-specific immunotherapy (AIT) aims to modulate the immune system to reduce the production or activity of Art v 1-specific IgE while increasing protective IgG4 antibodies (Source: PMID: 28024493).
Omalizumab binds to the Cε3 domain of IgE, preventing its interaction with the high-affinity IgE receptor (FcεRI) on mast cells and basophils. Allergen immunotherapy involves the administration of increasing doses of Art v 1 to induce immunological tolerance and shift the response from IgE-mediated to IgG4-mediated.
