Articular cartilage defects are localized areas of damage to the hyaline cartilage that covers the articulating surfaces of bones within synovial joints (StatPearls, 2023). These defects typically arise from acute traumatic injury, osteochondritis dissecans, or chronic mechanical wear, and they represent a significant clinical challenge because cartilage is avascular and has a very limited capacity for spontaneous repair (PubMed, PMID: 30213375). While 'articular cartilage defect' is a pathological condition rather than a specific molecular target, therapeutic interventions focus on restoring joint function and preventing the onset of osteoarthritis. Current pharmacological and biological approaches include the use of viscosupplements like hyaluronic acid to improve joint lubrication and corticosteroids to manage inflammation (NIH, 2022). Advanced regenerative techniques such as Matrix-induced Autologous Chondrocyte Implantation (MACI) are used to replace damaged tissue with functional chondrocytes. Research continues to explore molecular targets within the defect environment, such as matrix metalloproteinases (MMPs) and inflammatory cytokines, to improve the quality of repaired tissue. If left untreated, these lesions frequently progress to widespread joint degeneration and secondary osteoarthritis.
