Target intelligence / Profile preview

Aryl hydrocarbon receptor nuclear translocator (ARNT)

Target
ARNT
Molecular classification
Transcription factor, bHLH-PAS family (basic helix-loop-helix Per-ARNT-Sim), Receptor cofactor/partner (AHR dimerization partner)
01

Overview

Aryl hydrocarbon receptor nuclear translocator (ARNT, HIF-1β) is a bHLH-PAS transcription factor that heterodimerizes with ligand-activated AHR to bind xenobiotic response elements and induce genes in xenobiotic metabolism, and also serves as the beta subunit of HIF-1, partnering with HIF-1α to mediate hypoxia-responsive transcription; ARNT is essential for canonical AHR signaling, is implicated in cancer biology and hematopoiesis, and chromosomal translocations involving ARNT (e.g., TEL–ARNT) have been linked to acute myeloblastic leukemia.

Mechanism of action

AHR agonists: ligand binds AHR, AHR translocates to nucleus, heterodimerizes with ARNT (HIF1β), and the AHR–ARNT complex binds XREs to regulate target gene transcription (canonical AHR signaling); AHR antagonists/modulators: reduce AHR–ARNT-driven transcription by preventing AHR activation or dimerization, decreasing XRE binding and downstream gene expression; HIF pathway modulators: agents that alter HIF-α stability or activity affect ARNT-dependent HIF transcriptional complexes, modulating hypoxia-responsive genes.

03

Biological functions

Forms obligate heterodimers with ligand-activated Aryl hydrocarbon receptor (AHR) to drive transcription via xenobiotic response elements (XREs)Serves as the beta subunit of Hypoxia-inducible factor 1 (HIF-1), partnering with HIF-1α to mediate hypoxia-responsive gene expressionRegulates xenobiotic metabolism gene induction (e.g., CYP1A1) through AHR-ARNT complexesBroad transcriptional regulation via bHLH and PAS domains; can hetero- or homodimerize with other bHLH-PAS proteinsIn hematopoiesis, supports adult hematopoietic stem cell viability through regulation of BCL-2 and VEGF-A (functional role reported for ARNT)
04

Disease associations

Cancer (ARNT participates in AHR signaling that can promote or suppress tumor progression context-dependently; ARNT is also HIF-1β in hypoxia pathways relevant to tumor biology)Hematologic malignancy (TEL–ARNT fusion from t(1;12)(q21;p13) associated with acute myeloblastic leukemia)Hypoxia-related diseases via HIF signaling (ARNT as HIF-1β partner)
05

Safety considerations

Broad, pleiotropic transcriptional roles: targeting ARNT directly risks widespread effects on xenobiotic metabolism and hypoxia pathwaysPotential on-target toxicities via AHR pathway modulation (e.g., altered xenobiotic metabolism, immune modulation) since ARNT is required for canonical AHR signalingHematopoietic effects: ARNT is important for adult hematopoietic stem cell viability; perturbation could impact hematopoiesis
06

Interacting drugs

Indirect via AHR pathway: environmental ligands such as dioxin/TCDD and polycyclic aromatic hydrocarbons activate AHR, which then requires ARNT to signal
