Overview

ASB16 antisense RNA 1 (ASB16-AS1) is a cancer-associated long non-coding RNA (lncRNA), encoded on chromosome 17, affiliated with multiple aliases including C17orf65 and DKFZp762C2414. ASB16-AS1 has been implicated in the regulation of tumor cell proliferation, migration, and apoptosis through multiple mechanisms, such as acting as a competitive endogenous RNA (ceRNA) that sponges microRNAs including miR-760, modulating the expression of genes such as hepatoma-derived growth factor (HDGF), IGF1R, CDK6, and via protein interactions with HuR. Its dysregulation is linked to a variety of cancers, where it can act as an oncogene (e.g., in osteosarcoma) or tumor suppressor (e.g., in adrenocortical carcinoma). ASB16-AS1 expression levels are emerging as prognostic biomarkers in several malignancies. While targeting ASB16-AS1 remains preclinical, it is considered a potential therapeutic target for cancer therapy, with ongoing research seeking to clarify its value and mechanisms.

Mechanism of action

Functions as a miR-760 sponge to increase HDGF expression in osteosarcoma cells; Regulates IGF1R and CDK6 mRNA levels via interaction with RNA-binding protein HuR (ELAVL1) in adrenocortical carcinoma; Modulates the miR-185-5p/TEAD1 axis in colorectal cancer cells
