Overview

Asparaginyl-tRNA synthetase 1 (NARS1) is an essential cytoplasmic enzyme that catalyzes the covalent attachment of asparagine to its cognate tRNA (tRNA^Asn), a critical step in the process of translating genetic code into proteins[1][2][4]. NARS1 belongs to the Class II aminoacyl-tRNA synthetase family, characterized by a unique N-terminal extension (UNE-N), an anticodon-binding domain, and a C-terminal catalytic domain[3][4]. While its canonical function is the aminoacylation reaction enabling protein synthesis in all cell types, NARS1 also plays roles in cell signaling and is crucial for the proliferation of radial glial cells, especially during development of the cerebral cortex[1][2]. Mutations in NARS1 have been linked to developmental disorders including microcephaly, epilepsy, and neuropathy, and impair global protein synthesis[2][3][5]. There are currently no drugs known to target NARS1 directly, and its essential nature poses significant therapeutic challenges and safety concerns[1][2][4][5].

Mechanism of action

Because NARS1’s primary activity is enzymatic (aminoacylation of tRNAs), drugs would theoretically: - Inhibit its tRNA-charging activity, thus interfering with protein synthesis - Modulate enzyme stability/activity However, specific mechanisms from existing drug studies are not reported[5].
