Aspartate carbamoyltransferase (ATCase; EC 2.1.3.2) is a key enzyme in the de novo biosynthesis of pyrimidine nucleotides. It catalyzes the condensation of L-aspartate and carbamoyl phosphate to form N-carbamoyl-L-aspartate and inorganic phosphate, representing the first committed and rate-limiting step in pyrimidine biosynthesis. ATCase regulates cellular nucleotide pools and is allosterically regulated by CTP (inhibitor) and ATP (activator).
