Overview

Aspergillus fumigatus antigens encompass a heterogeneous group of molecular components derived from the saprophytic fungus Aspergillus fumigatus, including cell wall polysaccharides such as galactomannan and (1->3)-beta-D-glucan, as well as various secreted proteins and enzymes. These antigens play critical roles in fungal pathogenesis, including maintaining cell wall integrity, facilitating host tissue adhesion, and enabling the evasion of innate immune responses. In clinical practice, they serve as essential biomarkers for the diagnosis of invasive and allergic forms of aspergillosis through the detection of circulating antigens or specific host antibodies. Furthermore, these antigens are the functional targets for existing antifungal classes, such as echinocandins which disrupt cell wall synthesis, and are the focus of emerging immunotherapies, including monoclonal antibodies and chimeric antigen receptor (CAR) T cells, which aim to provide highly specific clearance of fungal hyphae.

Interventions targeting Aspergillus fumigatus antigens include the inhibition of fungal cell wall biosynthesis (e.g., via 1,3-beta-D-glucan synthase inhibition by echinocandins), disruption of fungal cell membrane integrity (e.g., via ergosterol binding by polyenes or inhibition of 14-alpha-demethylase by azoles), and direct recognition of fungal hyphae antigens by engineered immune cells like chimeric antigen receptor (CAR) T cells to induce cell lysis.
