Huangqi is the dried root of Astragalus membranaceus or Astragalus mongholicus, a fundamental medicinal herb in Traditional Chinese Medicine (TCM) used for centuries to treat fatigue and strengthen the body's immune defenses [1, 16]. It is not a single molecular target, but rather a complex natural product containing numerous bioactive constituents, including astragalosides (saponins), flavonoids, and diverse polysaccharides [5, 12]. These components exert broad pharmacological effects by interacting with multiple molecular pathways, such as the modulation of cytokine release (e.g., IL-6, TNF-alpha) and the activation of the PI3K/Akt/mTOR signaling pathway [8, 10]. Clinically, Huangqi is investigated for its potential in treating cardiovascular diseases, chronic kidney disease, and as an adjunctive therapy in cancer patients to mitigate the toxic effects of chemotherapy and enhance immune surveillance [13, 14]. It is widely recognized for its antioxidant and anti-inflammatory properties, which help protect various organs from oxidative stress [4, 16]. Because it is a multicomponent herbal remedy rather than a singular protein or receptor, 'Huangqi' is categorized as an incorrect entry for a specific therapeutic target molecule in the context of drug-receptor interactions [16].
Huangqi acts through a multi-target, multi-pathway mechanism. Key bioactive components such as astragalosides, flavonoids, and polysaccharides (APS) modulate the immune system by activating macrophages and T-cells, and regulating the Th1/Th2 cytokine balance [4, 5, 16]. It inhibits pro-inflammatory signaling pathways such as NF-kappaB and reduces the expression of cytokines like TNF-alpha and IL-6 [9, 10]. Additionally, Astragaloside IV, a primary saponin in Huangqi, is known to activate telomerase and stimulate the PI3K/Akt and Nrf2 pathways, providing potent antioxidant and cardioprotective effects [5, 13].
