Overview

Astringency is a sensory attribute marked by a drying, puckering feeling in the oral cavity, especially after ingestion of certain plant-derived compounds like tannins and polyphenols[3][4]. The perception of astringency results primarily from the interaction between these compounds and salivary proteins, particularly proline-rich proteins (PRPs). This leads to protein precipitation, aggregation, and alteration or depletion of the lubricating salivary film, exposing oral surfaces and increasing friction. Sensory perception is mediated by trigeminal nerves (responsible for tactile sensation) and taste nerves, and interpreted by mechanoreceptors and nociceptors rather than conventional receptor-ligand mechanisms[1][2][3]. There is no single molecule or receptor named "Astringency"; it is a multistage process involving complex oral physiology and biochemistry. Thus, "Astringency" cannot be mapped onto standard target definitions used for drug discovery, biomarker research, or molecular classification.
