Overview

AT-rich interactive domain-containing protein 2 (ARID2) is a DNA-binding subunit of the PBAF (poly-bromo-associated factor) SWI/SNF chromatin remodeling complex, which is essential for transcriptional regulation, chromatin architecture, and maintaining cellular identity. ARID2 contains a conserved AT-rich DNA interaction domain and zinc finger motifs that facilitate its binding to specific genomic regions. It plays crucial roles in embryonic patterning, cell cycle regulation, and tissue-specific gene activation. Loss-of-function mutations or deletions in ARID2 contribute to the development of cancers (notably hepatocellular carcinoma, lung cancer, and melanoma) by impairing normal chromatin remodeling and transcriptional control. Germline variants in ARID2 are causative for Coffin-Siris syndrome 6, which often presents with intellectual disability, autism spectrum disorder, ADHD, and congenital anomalies. ARID2-containing SWI/SNF complexes also play vital roles in osteoblast commitment and differentiation[1][2][3][4][5].
