Overview

AT-rich interactive domain-containing protein 3B (ARID3B) is a nuclear DNA-binding transcription factor belonging to the ARID family, characterized by an AT-rich interaction domain. ARID3B regulates chromatin structure and gene expression by binding specific AT-rich DNA sequences and interacting with key regulators of the cell cycle, such as E2F1 and the retinoblastoma protein (RB)[1][2][3][4]. It forms complexes with related protein ARID3A and is involved in both normal development and cancer biology, modulating cell proliferation, cell death, and “stemness” properties in both embryonic and cancer stem cells. Overexpression of ARID3B is implicated in the growth and malignant transformation of several cancers, notably neuroblastoma, ovarian cancer, and thyroid cancer, and is associated with an increase in CD133+ tumor stem cells[6][7]. Its role in cancer and stem cell biology positions it as a potential therapeutic target, though no targeted drugs are currently documented[1][2][4][6].
