ATP-binding cassette sub-family A member 8 (ABCA8) is a membrane-associated transporter belonging to the ABC transporter family, specifically the ABC1 subfamily, and is primarily involved in transporting organic anions, regulating lipid metabolism, and promoting cholesterol and sphingomyelin homeostasis[1][2][3][4][5]. ABCA8 is widely expressed, with particularly high levels in brain white matter, liver, heart, and skeletal muscle. In the central nervous system, it promotes sphingomyelin synthesis in oligodendrocytes, contributing to myelination. ABCA8 also mediates ATP-dependent transport of substances such as taurocholate, estrone sulfate, and ochratoxin A, with its roles spanning xenobiotic clearance and lipid trafficking. Disease associations include several cancers (often as a tumor suppressor gene), myelination disorders, and certain lipid metabolism disorders. While not currently a drug target, its activity influences chemoresistance and tumor progression in some contexts[3].
