ATP-binding cassette sub-family A member 9 (ABCA9) is a member of the ATP-binding cassette (ABC) transporter superfamily, specifically the ABCA (ABC1) subfamily, and is encoded by the ABCA9 gene in humans[1][2][3][5][7]. The protein is a full-size ABC transporter, containing two transmembrane domains and two nucleotide-binding domains, consistent with proteins that transport various molecules across extra- and intracellular membranes. ABCA9 is ubiquitously expressed, with highest mRNA levels in the heart, brain, and fetal tissues; it is upregulated during monocyte differentiation into macrophages and suppressed by cholesterol import[2][3]. Functionally, ABCA9 participates in lipid transport and cellular lipid homeostasis, especially in macrophages, and is part of a cholesterol-responsive regulatory network[2][3][5][7]. Its expression and genetic variants have been implicated as biomarkers in several cancers, and its altered function may play a role in rare congenital disorders and metabolic disease[3][7]. No specific drugs are currently known to interact directly with ABCA9, and its safety profile as a therapeutic target remains uncharacterized.
