ABCC8 encodes the sulfonylurea receptor 1 (SUR1), a regulatory subunit of the ATP-sensitive potassium channel (KATP channel) in pancreatic beta cells. The KATP channel is crucial for coupling cell metabolism to electrical activity and insulin secretion. Mutations in ABCC8 can cause congenital hyperinsulinism, neonatal diabetes, and other disorders of glucose homeostasis.
Sulfonylureas bind to SUR1, closing the KATP channel and stimulating insulin release.
