Overview

ABCC8 (SUR1) is the regulatory subunit of the ATP-sensitive potassium (KATP) channel, primarily found in pancreatic beta cells. It plays a crucial role in glucose-stimulated insulin secretion by controlling membrane potential in response to intracellular ATP/ADP levels. Mutations in ABCC8 are associated with congenital hyperinsulinism and permanent neonatal diabetes mellitus. It is also a target for sulfonylurea drugs used in the treatment of type 2 diabetes.

Mechanism of action

Sulfonylureas bind to SUR1, causing closure of KATP channels, which leads to membrane depolarization and increased insulin secretion.
