ATP-dependent RNA helicase DDX51 is an enzyme from the DEAD-box family involved in the processing and maturation of ribosomal RNAs, particularly the 28S rRNA, and the assembly of the 60S ribosomal subunit. Like other DEAD-box helicases, DDX51 acts by remodeling RNA structures or RNA–protein interactions through ATP hydrolysis, facilitating critical steps in RNA metabolism. DDX51 is ubiquitously expressed and is essential for ribosomal assembly and overall cell physiology. Elevated DDX51 expression has been reported in cancers such as esophageal squamous cell carcinoma, where it promotes cell proliferation, migration, invasion, and is correlated with poor clinical outcomes; it exerts part of its oncogenic effect by activating the PI3K/AKT signaling pathway. Currently, no drugs are known to specifically target DDX51, but as a member of the broader DEAD-box helicase family, it represents an emerging area of therapeutic research.
