Overview

The ATP-sensitive potassium channel regulatory subunit SUR1 is a regulatory component of the K_ATP channel, an octameric complex of four Kir6.2 and four SUR1 subunits. It regulates the K_ATP channel's activity by linking cellular metabolism to membrane excitability. SUR1 is the molecular target of sulfonylurea drugs used in type 2 diabetes. Mutations in ABCC8/SUR1 can cause congenital hyperinsulinism or neonatal diabetes. Additionally, SUR1 plays a role in cardiac tissue under stress.

Mechanism of action

Sulfonylureas and glinides bind to SUR1, causing closure of K_ATP channels in pancreatic beta cells, leading to depolarization and insulin release.
