Overview

Kir6.1 is a pore-forming subunit of the ATP-sensitive potassium (KATP) channel, encoded by the KCNJ8 gene. KATP channels are hetero-octameric complexes composed of four Kir6.x subunits (Kir6.1 or Kir6.2) and four regulatory sulfonylurea receptor (SUR) subunits. These channels link cellular metabolism to membrane excitability by sensing intracellular nucleotide concentrations and modulating potassium ion flow. Kir6.1 is highly expressed in vascular smooth muscle, brain pericytes, skeletal muscle, cardiac tissue, pancreatic β-cells, and neurons. It plays a crucial role in regulating vascular tone, blood pressure, cerebral perfusion, and protecting against metabolic stress.

Mechanism of action

Sulfonylureas inhibit KATP channels by binding to the SUR subunit, leading to membrane depolarization and increased insulin secretion in pancreatic beta cells.
