ATP synthase subunit OSCP (Oligomycin Sensitivity Conferring Protein) is a key component of the mitochondrial F-type ATP synthase (Complex V). It is part of the peripheral stalk and connects the catalytic core (α3β3) in the F1 domain to components in the Fo domain, stabilizing the complex during rotary catalysis. The OSCP subunit acts as a stator element that prevents rotation of certain parts of ATP synthase while allowing other parts to rotate freely during catalysis, enabling efficient coupling between proton translocation through Fo and conformational changes in F1 necessary for ATP synthesis or hydrolysis. It is encoded by the ATP5O gene.
Inhibits ATP synthase by binding to the Fo subunit, blocking proton flow.
