ATPase phospholipid transporting 11B (ATP11B) is a member of the P-type ATPase family, which are membrane-bound enzymes that utilize ATP hydrolysis to transport phospholipids, notably driving the translocation (flipping) of phosphatidylserine and other aminophospholipids from the outer to the inner leaflet of biological membranes[1][2][3][6]. This action is essential for maintaining the lipid asymmetry in the plasma membrane, which impacts cell signaling, apoptosis, vesicle formation, and membrane dynamics. The gene ATP11B encodes this protein in humans and is expressed in a broad variety of tissues. Although many P-type ATPases are well-characterized, ATP11B remains a "putative" or probable transporter based on sequence homology and predicted function. Its expression and proper activity are critical for cellular homeostasis, and its dysregulation has been implicated in cancer biology and possibly other diseases, but there are currently no approved drugs directly targeting ATP11B as of September 2025[1][2][5][6].
