Atrial natriuretic factor receptor A (NPR-A), also known as guanylyl cyclase-A (GC-A), is a transmembrane receptor that binds atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP). Upon ligand binding, NPR-A catalyzes the production of cyclic GMP (cGMP), leading to vasodilation, natriuresis, diuresis, and inhibition of cardiac hypertrophy. NPR-A is a therapeutic target for hypertension and heart failure, but clinical use of NPR-A agonists is limited due to safety concerns.
ANP/BNP binding increases intracellular cGMP levels, activating downstream pathways such as PKG.
