The cochlear nerve is a branch of the vestibulocochlear nerve (cranial nerve VIII), a sensory cranial nerve responsible for carrying auditory information from the cochlea in the inner ear to the cochlear nuclei in the brainstem[1][2][3][5]. It consists of afferent fibers originating from bipolar neurons in the spiral (cochlear) ganglion, which synapse with the hair cells of the organ of Corti. Most fibers are large, myelinated type I neurons (projecting to inner hair cells), with a minority being small, unmyelinated type II neurons (projecting to outer hair cells)[1][2][3]. The cochlear nerve transmits electrical impulses, generated from hair cell activation by sound waves, to the brain where these signals are interpreted as hearing. Damage to the cochlear nerve from trauma, tumors (such as acoustic neuroma), infections, or demyelinating diseases can result in sensorineural hearing loss, tinnitus, or additional auditory neuropathies[1][3][5]. As a nerve rather than a molecular target, it does not directly act as a therapeutic receptor, enzyme, or transporter but may be the object of device-based interventions (e.g., cochlear implants) rather than pharmacological targeting[1][2][3][5]. Key points: - The cochlear nerve is not a molecule, receptor, enzyme, or transporter, but a sensory nerve/fiber bundle. - It is not a conventional "therapeutic target" for drugs; thus most database frameworks for molecular drug targets will flag this as an inappropriate entry[1][3][5]. - It is relevant in neuroanatomy and clinical discussions on hearing loss and certain surgical interventions, but not in the context of drug-target interactions.
