Overview

The Augmin complex, also known as the HAUS complex in humans, is a critical eight-subunit protein assembly essential for spindle microtubule organization during mitosis (Goshima et al., 2008; Uehara et al., 2009). Its primary biological function is to facilitate branching microtubule nucleation by recruiting the gamma-tubulin ring complex (gamma-TuRC) to the sides of pre-existing microtubules (Petry et al., 2013). This process significantly increases microtubule density within the mitotic spindle, ensuring robust chromosome capture and alignment (Lawo et al., 2009). By generating microtubules from within the spindle rather than just the centrosomes, the complex maintains spindle structural integrity and facilitates efficient cell division (Meireles et al., 2020). In the context of disease, the Augmin complex is investigated as a potential therapeutic target in oncology, as its disruption leads to spindle multipolarity, chromosome misalignment, and subsequent mitotic catastrophe in cancer cells (Song et al., 2018). While no specific small-molecule inhibitors are currently in clinical use, the complex represents a selective vulnerability in tumors with centrosome amplification or specific mitotic dependencies.
