Auricular points are specific anatomical locations on the external ear (auricle) utilized in auriculotherapy and acupuncture to treat a variety of physical and psychological conditions [NIH NCCIH, Acupuncture: In Depth]. These points are organized according to a somatotopic map, where the ear is viewed as an inverted fetus representing the entire body [Hou et al., 2015, Evidence-Based Complementary and Alternative Medicine]. The therapeutic mechanism is primarily attributed to the stimulation of cranial nerves, most notably the auricular branch of the vagus nerve (ABVN), which provides a direct pathway to the central nervous system and modulates autonomic activity [Butt et al., 2020, Journal of Anatomy]. Clinical applications include the management of chronic pain, substance withdrawal, and anxiety [Zhao et al., 2015, Medical Acupuncture]. However, auricular points are not molecular targets such as receptors, enzymes, or transporters; they are physiological reflex points and do not have direct interactions with pharmacological drugs [StatPearls, Acupuncture]. Stimulation is typically achieved through the use of needles, seeds, or transcutaneous electrical nerve stimulation (TENS). Research suggests that activating these points can influence the release of endogenous opioids and neurotransmitters like serotonin and dopamine. Despite their clinical use, they lack the defined biochemical structure required for traditional drug-target classification.
Stimulation of auricular points triggers neural pathways, primarily involving the auricular branch of the vagus nerve (ABVN), which projects to the nucleus tractus solitarius and modulates the central nervous system and autonomic functions [Hou et al., 2015, Evidence-Based Complementary and Alternative Medicine; Butt et al., 2020, Journal of Anatomy].
