Overview

Autologous keratinocytes are a patient's own skin cells harvested, expanded in vitro, and grafted onto damaged skin to restore the epidermis. This method is used for treating extensive burns, chronic wounds, and certain genetic or pigmentary skin disorders. The keratinocytes, particularly those with stem cell-like properties, reconstitute a functional stratified epithelium after transplantation. EGFR signaling, integrins, and Wnt signaling are key pathways involved in successful regeneration. The grafts may contain keratinocytes, melanocytes, and sometimes fibroblasts. Advantages include low rejection risk and the ability to cover large areas from small biopsies. Limitations include the need for specialized lab facilities and a time lag for preparation.
