Autophagy machinery refers to the collection of proteins and complexes that orchestrate autophagy, a conserved cellular process responsible for degrading and recycling cytoplasmic components through lysosome-dependent pathways. This machinery is essential for maintaining cellular homeostasis, responding to stress, and regulating development, immunity, and cell death. The core machinery includes the ULK1 complex (initiation), the PI3K complex (nucleation), ATG9 vesicles and the ATG12-ATG5-ATG16L1 and LC3/GABARAP conjugation systems (elongation), and lysosomal hydrolases (degradation). Autophagy is regulated by nutrient-sensing kinases like mTOR and AMPK. Defects in the pathway are implicated in various diseases, including neurodegeneration and cancer, making it a therapeutic target.
