Overview

Autophagy-related protein 14 (ATG14) is a key regulatory subunit of the autophagy-specific Class III phosphatidylinositol 3-kinase (PI3KC3) complex, which includes VPS34 and Beclin 1[1][2]. ATG14 directs the complex to the site of autophagosome formation (in mammals, ER-associated membranes), serving as a connector and membrane-targeting factor through its N-terminal and C-terminal regions, respectively. The C-terminal BATS domain preferentially binds highly curved phosphatidylinositol 3-phosphate–containing membranes, targeting the PI3KC3 complex to nascent autophagosomal membranes[1][2]. ATG14’s interaction with Beclin 1 and VPS34 is essential for initiation of autophagy. Its function is fundamental for cellular homeostasis, and dysregulation has been implicated in cancer, neurodegenerative diseases, infections, and inflammatory conditions. While ATG14 itself is not directly targeted by approved drugs, its central role in autophagy regulation makes it an important therapeutic target for autophagy-modulating interventions[2][1].
