Axin2, also known as Conductin or Axil, is a scaffolding protein and a core component of the β-catenin destruction complex, which negatively regulates the canonical Wnt signaling pathway. It promotes the phosphorylation and subsequent degradation of β-catenin, thereby limiting Wnt pathway activity. Axin2 is induced by Wnt signaling, likely forming a negative feedback loop to fine-tune Wnt responses. It interacts with APC, GSK3, CK1, and β-catenin to coordinate the sequential phosphorylation of β-catenin, marking it for degradation. Mutations in the AXIN2 gene are associated with colorectal cancer, indicating its role as a tumor suppressor. Structurally, Axin2 contains a GSK3-binding domain (GBD) that mediates interactions with the E3 ubiquitin ligase SIAH1, which regulates Axin2 degradation. Although Axin1 and Axin2 perform similar functions, their expression patterns and some molecular interactions differ, suggesting non-redundant roles in certain contexts. The target name "Long noncoding RNA A33/Axin2/β-catenin" is incorrect; Axin2 is a protein-coding gene, not a long noncoding RNA.
