AXL is a receptor tyrosine kinase belonging to the TAM (TYRO3, AXL, MER) family. It is activated by its ligand GAS6, leading to receptor dimerization, autophosphorylation, and activation of downstream signaling pathways including PI3K/AKT/mTOR, RAS/MAPK, NF-κB, and JAK/STAT. AXL plays a crucial role in cell proliferation, survival, migration, invasion, and immune modulation, and is frequently overexpressed in various cancers, where it promotes tumorigenesis and drug resistance. Targeting the Gas6/AXL axis is being explored as a therapeutic strategy in cancer.
