B and T lymphocyte attenuator (BTLA), also known as CD272, is an immunomodulatory receptor belonging to the CD28 immunoglobulin superfamily. It is widely expressed on immune cells and functions as a co-inhibitory receptor, downregulating immune responses. BTLA interacts with HVEM, leading to ITIM/ITSM phosphorylation and recruitment of phosphatases like SHP-1/SHP-2, which inhibit lymphocyte activation. It plays roles in immune tolerance, infection, and innate immune suppression, making it a potential therapeutic target, particularly in cancer immunotherapy and autoimmune diseases.
