The BAFF receptor (BAFF-R/TNFRSF13C) is an essential surface molecule on maturing/mature human B lymphocytes that mediates critical pro-survival signals upon engagement with its unique ligand—BAFF. It orchestrates downstream pathways such as NF-kappaB2 and PI3K/AKT/mTOR that collectively ensure proper development, metabolic fitness, longevity, differentiation potential—and thus effective humoral immunity—of peripheral naive/memory/plasma cell populations. Dysfunctional signaling through this axis underlies several immunodeficiencies as well as autoimmunity when dysregulated.
