Target intelligence / Profile preview
B cell differentiation refers to the biological process by which immature B lymphocytes mature into antibody-secreting plasma cells or memory B cells, a critical step in humoral immunity. This process involves activation by antigens via the B cell receptor (BCR), often with T cell help through cytokines like IL-4, IL-21, and CD40L interactions, leading to proliferation, class-switch recombination, and somatic hypermutation in germinal centers. Key factors include B cell growth factors (BCGFs, such as BCGF I/IL-4) that drive cell cycle progression and B cell differentiation factors (BCDFs) that promote maturation to immunoglobulin-secreting cells. Transcription factors like Blimp-1 are essential for terminating the B cell program and initiating plasma cell identity. Dysregulation of B cell differentiation contributes to diseases like diffuse large B-cell lymphoma (DLBCL), where molecular subtypes (e.g., germinal center B-cell-like or activated B-cell-like) reflect arrested differentiation stages and influence prognosis. While not a specific molecular entity, this process is targeted indirectly via cytokines or pathways in immunotherapy, but lacks direct small-molecule drugs due to its complexity as a cellular pathway rather than a single protein.
