B-cell lymphoma 9 protein (BCL9) is a transcriptional co-activator involved in the Wnt/β-catenin signaling pathway. It plays a critical role in regulating gene expression related to cell growth, proliferation, and differentiation. BCL9 acts as a scaffolding component of the Wnt enhanceosome, facilitating the interaction between β-catenin and TCF/LEF transcription factors to activate specific target genes. Overexpression or chromosomal aberrations involving BCL9 are associated with various malignancies, including acute lymphoblastic leukemia (ALL) and colorectal cancer. Common genetic variations within the distal region of the *BCL9* gene confer risk for schizophrenia and may be associated with bipolar disorder and major depressive disorder. Targeting the β-catenin/BCL9 interaction represents an attractive therapeutic approach.
