B-cell lymphoma-extra large protein (Bcl-xL) is an anti-apoptotic member of the Bcl-2 protein family. It is encoded by the BCL2-like 1 gene and primarily resides in the mitochondrial outer membrane. Bcl-xL prevents apoptosis by inhibiting the release of cytochrome c from the mitochondria, thereby blocking caspase activation. Overexpression of Bcl-xL is associated with tumor progression and chemoresistance in various cancers, making it a target for therapeutic intervention. Beyond apoptosis, Bcl-xL is also involved in autophagy, neuronal survival, calcium signaling, and cellular senescence.
