B-cell receptors (BCRs) specific for Adeno-associated virus serotype 2 (AAV2) capsid epitopes and model antigens like SIINFEKL are critical mediators of the humoral immune response in gene therapy contexts. These receptors are membrane-bound immunoglobulins that recognize viral capsid proteins, triggering B-cell activation and the subsequent production of neutralizing antibodies (NAbs) that can severely limit the efficacy of AAV-mediated gene delivery (Mingozzi & High, 2013, Nature Reviews Genetics). Therapeutic strategies include the use of B-cell depleting agents like rituximab or the development of tolerogenic nanoparticles designed to specifically silence B cells expressing these receptors. Successfully managing these BCR-mediated responses is essential for the safety and durability of genetic medicines.
