Overview

B-cell receptors (BCRs) specific for HIV-1 envelope (Env) epitopes are the primary molecular targets for modern HIV-1 vaccine design, particularly germline-targeting strategies (Jardine et al., 2013, Science). These receptors are membrane-bound immunoglobulins on B cells that recognize the HIV-1 Env glycoprotein, the only viral protein exposed on the virion surface (Burton & Hangartner, 2016, Annu Rev Immunol). In the context of HIV-1 infection, the virus's high mutation rate and glycan shield typically prevent these BCRs from evolving into broadly neutralizing antibodies (bNAbs). Therapeutic interventions, such as the engineered immunogen eOD-GT8 60mer, are designed to bind and activate rare germline B cells with the potential to develop into bNAbs (Leggat et al., 2022, Science). By specifically engaging these BCRs, these therapies aim to guide the immune system through somatic hypermutation and affinity maturation to produce a robust protective response against diverse HIV-1 strains (Haynes et al., 2023, Nat Biotechnol).
