Overview

B-cell receptors (BCRs) and antibodies specific for Ag85B and ESAT-6 are essential components of the adaptive immune system's response to Mycobacterium tuberculosis (Mtb). Ag85B (Antigen 85B) is a mycolyltransferase essential for cell wall synthesis, while ESAT-6 (Early Secretory Antigenic Target-6) is a major virulence factor secreted by the Mtb ESX-1 system (UniProt: P9WQP3, P9WNK5). BCRs on the surface of B-cells recognize these antigens, leading to the production of specific antibodies (IgG, IgA, IgM) that contribute to host defense through opsonization, neutralization, and modulation of cellular immunity (PubMed: 28932019). These immune components are the primary targets of several tuberculosis subunit vaccines, such as H56:IC31 and H1:IC31, which aim to induce protective humoral and B-cell memory (PubMed: 21844364). Additionally, the detection and quantification of these antibodies serve as important biomarkers for diagnosing active tuberculosis and evaluating the immunogenicity of vaccine candidates in clinical trials (PubMed: 15939750).
