Overview

B cell scaffold protein with ankyrin repeats 1 (BANK1) is a B cell-specific adaptor protein that plays a critical role in integrating signals downstream of the B cell receptor (BCR) and Toll-like receptors (TLRs)[1][2][5][6]. BANK1 interacts with key signaling proteins (BLK, LYN, MyD88, TRAF6, PLCγ2), influencing cytokine production, cell activation, and the regulation of B cell memory responses[2][5]. Genetic variants in BANK1 alter its regulatory domains and expression, leading to increased risk for autoimmune diseases such as systemic lupus erythematosus, rheumatoid arthritis, and systemic sclerosis[2][5][6]. BANK1 is being explored as a biomarker in autoimmune conditions and cancer but is not yet a clinical therapeutic target[3][5][6].
