Overview

B melanoma antigen 1 (BAGE1) is a member of the BAGE family of cancer-testis antigens, which are proteins typically expressed only in the germ cells of the testis and silenced in normal adult somatic tissues. However, BAGE1 is frequently re-expressed in various malignancies, including approximately 22% of melanomas, as well as bladder and lung carcinomas, making it a highly specific target for cancer immunotherapy. Because the testis is an immunologically privileged site lacking HLA Class I expression, BAGE-specific T-cells do not attack normal testicular tissue, providing a wide therapeutic window. In clinical development, BAGE1 has been targeted primarily through peptide-based vaccines and adoptive T-cell therapies designed to stimulate the immune system to recognize and destroy BAGE-positive tumor cells. Its role as a tumor-specific antigen makes it a valuable candidate for precision oncology and multi-antigen vaccine strategies.
